Palcic, Monica M Professor Our research is in the area of mechanistic enzymology with a goal of obtaining a detailed description, on the molecular level, of enzyme reactions. Two different types of enzyme are currently under investigation, amine oxidases and enzymes involved in oligosaccharide biosynthesis, both glycosyltransferases and glycosidases. Complex carbohydrates covalently linked to proteins and lipids are found on virtually all mammalian cell surfaces. These molecules are biosynthesized by glycosyltransferases that catalyze the sequential addition of a monosaccharide unit from an activated nucleotide donor to an acceptor. While changes in the structures of cell surface carbohydrates occur in both normal and aberrant cellular development, the underlying biological significance of these changes is unclear. To better understand the functional role of cell surface carbohydrates, our research is directed towards the design and evaluation of inhibitors of glycosyltransferases and glycosidases on purified enzymes and cell culture systems and developing improved assays, primarily capillary electrophoresis methods. This requires enzymatic synthesis of oligosaccharides for biological evaluations as shown below.
